استخراج و شناسایی ترکیبات فیتواستروژنی، الاجیک، و سیرینژیک اسید از پوست انار

چکیده

در این تحقیق از پوست تازۀ سه رقم انار معروف ایرانی توسط چهار حلال متفاوت به روش سوکسله عصاره‏گیری شد. عصاره‏ها برای تشخیص کمّی و کیفی دو نوع تانیک‌اسید و هفت نوع فیتو‏استروژن به دستگاه کروماتوگرافی مایع با کارایی بالا (HPLC) جفت‌شده با شناساگر مرئی‌ـ‌فرابنفش (UV-Vis) تزریق شدند. همچنین در پژوهش انجام‌شده راندمان استخراج از پوست سه رقم انار و چهار حلال با یکدیگر مقایسه شدند. کمّیت و کیفیت ترکیبات بررسی‌شده در عصاره‏ها ازطریق مقایسۀ زمان بازداری استانداردهای متناظر ترکیبات و با روش تزریق استاندارد خارجی به دستگاه مشخص شدند. نتایج آزمایش‌ها نشان داد در میان حلال‏ها مخلوط مساوی از چهار حلال آب، اتانول، استون، و اتیل‏استات و در بین ارقام انار رقم پوست‌گلی ملس ساوه بیشترین میانگین راندمان استخراج را داشته‌اند. نتایج آزمایش‌های دستگاه کروماتوگرافی بیانگر این است که در بین هفت فیتو‏استروژن فقط سه فیتو‏استروژن استریول (تا 45/7 میلی‌گرم بر 100 گرم پوست تازه)، تستسترون، و استرادیول (تا 43/8 میلی‌گرم در 100 گرم پوست تازه) در مقدار قابل قبول تقریباَ در تمامی نمونه‏ها شناسایی شدند. در بین دو نوع تانیک‌اسید، الاجیک‌اسید در کمیّتی بسیار بالا (تا 0/103 میلی‌گرم در 100 گرم پوست تازه)، و در تمامی عصاره‏ها و سیرینژیک‌اسید نیز در اغلب نمونه‏ها شناسایی شدند.

چکیده [English]

In this work, fresh pomegranate peel of three dominant Iranian cultivaris was extracted by soxhlet extraction method via different four solvents. The extracts were analyzed by high performance liquid chromatography (HPLC), using ultraviolet-visible spectrometric detection. In this study, the extraction yields and presence of seven phytoestrogenic compounds, two tannic acids and their quantities in extracts were evaluated. The compounds were identified and quantified according to their retention times with external standards. Comparison of fresh pomegranate peel of three cultivars indicated that yield of extraction from PoostGoliyeMalaseSaveh variety was more than other two cultivars and the yield of extraction of extracts obtained from the mix solvent was more than three other solvents (p≤0.05). HPLC analysis indicated besides ellagic and syringic acids, some of phytoestrogens such as Esteriol (up to 7.45 mg/100 g fresh peel), Testosteron and α-Estradiol (up to 8.43 mg/100 g fresh peel) were also found in extract of pomegranate peels. The results show that, between two tannic acids, high value of ellagic acid (up to 103/0 mg/100g fresh peel of post siyahe-Ardestan) were recognized in all extracts.
