مقایسه کاربرد ستون‌های مختلف برای آنالیز آکریلامید در چیپس سیب زمینی با روش کروماتوگرافی مایع

چکیده

آکریلامید ترکیبی است سمی که در برخی از مواد غذایی فرآیند شده ایجاد می­­­­­­­­­­­شود. هدف از این پژوهش، ارائه یک روش آسان و قابل اطمینان برای اندازه­گیری آکریلامید در چیپس سیب زمینی بود. بدین منظور، اثر 5 ستون ODS-H-C18 Optimal، Sphere-Image 80-5 ODS 2، Zorbax Hilic Plus، Lichrosorb-100 NH2 و Shodex NH2P-50 4E بر جداسازی و اندازه­گیری آکریلامید در چیپس بررسی گردید. بهترین نتایج با کاربرد ستون آمینه نسل جدید Shodex NH2P-50 4E، به دست آمد و پیک آکریلامید با موفقیت و بدون نیاز به انجام مراحل پاکسازی با فاز جامد جداسازی شد. منحنی کالیبراسیون استاندارد در محدوده μg/L 5000-100 کاملا خطی بود (997/0=R2). درصد بازیافت در محدوده 9/95% تا 2/102% ، حد شناسایی و حد تعیین مقدار کمی نیز به ترتیب  μg/kg9 و μg/kg 27 به دست آمد. تکرار پذیری این روش مناسب بوده و شاخص انحراف استاندارد نسبی برابر 39/4% بود.

Acrylamide is a toxic compound produced in some processed foods. The purpose of this research was to provide an easy and reliable method for measuring acrylamide in potato chips by HPLC-PDA. For this purpose, the effect of 5 columns ODS-H-C18 Optimal, Sphere-Image 80-5 ODS 2, Zorbax Hilic Plus, Lichrosorb-100 NH2 and Shodex NH2P-50 4E were investigated on the separation and measurement of acrylamide in chips. The best results were obtained with the use of the new generation of Shodex NH2P-50 4E amino columns, and acrylamide peak was isolated successfully without the need for solid state purification steps. The standard calibration curve was linear in the range of 100-5000 μg / L (R2 = 0.997). Recycling percentages in the range of 95.9% to 102.2%, the detection limit and the limit of value were 9 and 27 μg / Kg respectively. The repeatability of this method was appropriate and the relative standard deviation was 4.39%.
